When to Take Magnesium Glycinate: What the Research Shows

Almost every magnesium glycinate on the Australian market tells you to take it in the evening. Ours does. Very few of them explain why, and the honest answer is more interesting than the label suggests.
This is what the research actually shows about timing. How long it takes for a dose to reach your bloodstream, what the trials that reported an effect on sleep actually did, whether magnesium is sedating in the way people assume, and the one timing decision that has real evidence behind it. I have kept the claims to what the studies measured.

The short answer
Follow the directions on your product. For most magnesium glycinate in Australia, including ours, that means a two capsule serve in the evening, taken daily rather than now and then.
If you want a reason rather than an instruction, the best available one comes from the trials themselves. The most recent randomised controlled trial on magnesium bisglycinate had its participants take the dose 30 to 60 minutes before bed, so an evening serve is at least consistent with how the research was conducted.
What the evidence does not support is the idea that the hour on the clock is doing heavy lifting. Magnesium is a nutrient, not a drug with a dose response curve you can feel. The sections below set out why.
What happens in the hours after you swallow it
This is the part that reframes the whole question. Magnesium does not arrive quickly.
A 2026 randomised, open label pharmacokinetic study published in Scientific Reports gave 24 healthy volunteers either 750 mg or 2000 mg of oral magnesium oxide and took blood samples for 12 hours afterwards. At the lower dose, the median time to peak serum magnesium was six hours.
At the 750 mg dose, serum magnesium took a median of six hours to reach its peak. Tripling the dose brought that forward to two hours, but did not produce a proportionally larger rise in blood magnesium.
Two caveats worth stating plainly. That study used magnesium oxide, not glycinate, and all 24 participants were male. So the exact numbers do not transfer directly to a glycinate capsule taken by a woman. What does transfer is the shape of the curve: an oral magnesium dose moves into the blood over a matter of hours, not minutes.
That has a practical consequence. If you take a capsule and expect to notice something within twenty minutes, you are working to a timeline the biology does not run on. It also means a serve taken at 6pm and a serve taken at 9pm are not meaningfully different events by the time your body has dealt with them.
Why almost every label says evening
Three reasons, and they are not equally good.
It matches how the trials were run. The randomised, double blind, placebo controlled trial published in Nature and Science of Sleep gave 155 adults aged 18 to 65 two capsules of 893 mg magnesium bisglycinate, delivering 250 mg of magnesium and 1523 mg of glycine, and instructed them to take it 30 to 60 minutes before bedtime for four weeks. Insomnia Severity Index scores fell by 3.9 points in the magnesium group against 2.3 in the placebo group, a statistically significant difference at p equals 0.049 with a small effect size of d equals 0.2. If you want to copy a protocol that produced a measured result, that is the protocol.
There is a genuine circadian story underneath it. A 2016 paper in Nature found that the concentration of magnesium ions inside cells rises and falls on a daily cycle, and that this oscillation acts as a cell autonomous timekeeping component. The researchers saw it in human cells, in mouse fibroblasts, in a fungus and in a single celled alga, organisms separated by more than a billion years of evolution. Magnesium is not simply a passenger in the cell's daily rhythm.
Being careful here: that is cell biology, not a dosing instruction. Nobody has shown that timing a supplement to that rhythm changes an outcome in a person. It is a reason the question is worth asking, not an answer to it.
Habit and convenience. The least scientific reason is probably the most powerful one. Evening is a reliable cue. People who tie a supplement to brushing their teeth take it more consistently than people who mean to take it at some point during the day, and with a nutrient, consistency is the thing that actually accumulates.
Does magnesium make you sleepy?
Not in the way a sedative does, and it is worth being clear about that before you build an evening around it.
A 2025 review in Nature and Science of Sleep sets out the mechanisms that have been proposed. Magnesium ions interact with GABA receptors in a way that dampens neural excitability, and magnesium inhibits the NMDA receptor, which lowers calcium concentration inside muscle cells. The review also notes magnesium's role in the enzyme pathway that makes melatonin, and its regulation of the cellular clock. At no point does it describe magnesium as a sedative, and neither should anyone selling it to you.
The trial evidence matches that modesty. In the 2012 double blind placebo controlled trial in the Journal of Research in Medical Sciences, 43 older adults with primary insomnia took 500 mg of elemental magnesium daily as magnesium oxide for eight weeks. Sleep onset latency shifted from 1.3 hours to 1.1 hours, and Insomnia Severity Index scores moved from 16.5 to 14.1. Serum melatonin rose slightly and cortisol fell slightly. Real, measurable, and a long way from switching off a light.
Two things to notice about that trial. The dose was 500 mg of elemental magnesium, which sits well above the 350 mg daily upper level the Australian and New Zealand Nutrient Reference Values set for supplemental magnesium. And the participants were aged 60 to 75 and had a diagnosed sleep disorder, so they are not a stand in for a healthy 34 year old. With food or without
This one is genuinely unsettled, and anyone who tells you otherwise is filling a gap in the literature with confidence.
What is established is that magnesium crosses the gut wall two ways. Jahnen-Dechent and Ketteler in Clinical Kidney Journal describe the bulk of it moving passively between cells, with a smaller regulated route through the TRPM6 and TRPM7 channels that works hardest when your magnesium is low. Food slows gastric emptying, which spreads the dose out, and for some people that means a gentler experience.

When splitting the dose makes sense
Here is the one timing decision with solid evidence behind it, and it has nothing to do with the clock.
The classic measurement, from a 1991 study published in the Journal of Clinical Investigation, fed healthy volunteers five different magnesium loads and measured what got in. The proportion absorbed fell steadily as the dose rose, from 65 per cent at the lowest intake down to 11 per cent at the highest. The 2026 pharmacokinetic study points the same way from a different angle: tripling the dose did not produce a proportionally bigger rise in serum magnesium.
So the rule is about dose size, not hour. Two smaller serves are generally better absorbed and better tolerated than one large one. If a product hands you 400 mg of elemental magnesium in a single hit, splitting it morning and evening is the sensible move. At 100 to 200 mg of elemental magnesium, which is where most well formulated glycinate products sit, there is nothing to split.
Whatever is left unabsorbed stays in the gut and draws water into it, which is the entire mechanism behind magnesium's reputation for digestive upset. Poor absorption is not a neutral trait. The form comparison is in our guide to magnesium forms.
The part that matters more than the hour
Magnesium intake is a daily number, not an event. The Nutrient Reference Values set the recommended dietary intake at 310 mg a day for women aged 19 to 30 and 320 mg from 31 onwards, from all sources, with most of it meant to come from food. A supplement is topping up a running total.
There is no loading phase, nothing to feel in the first hour, and no reason to save it for the nights you think you will need it. Four consistent nights and three missed ones is a lower intake than seven consistent ones, and that is the whole of the arithmetic.
Which is why the best time to take magnesium glycinate is, unromantically, the time you will actually remember.
The bottom line
Evening is the conventional instruction, it matches the protocol used in the most recent randomised trial, and there is real cell biology showing magnesium moves on a daily rhythm. None of that adds up to a specific magic hour, and a dose takes hours rather than minutes to reach peak concentration in the blood.
The two decisions with actual evidence behind them are dose size, because absorption falls sharply as the dose climbs, and consistency, because intake is a daily figure. Get those right and the clock takes care of itself.
