Magnesium Glycinate: What It Actually Is and How People Use It

Magnesium glycinate has gone from a specialist ingredient to the most requested form of magnesium in Australian pharmacies in about five years. Most people who buy it can tell you it is the gentle one. Far fewer could tell you what it actually is.

This is the plain version. What the molecule is, what magnesium does in the body, what the glycine half is doing there, and what the research shows about how much of it you absorb. No claims about what it will do for you, because that is not what the evidence supports and it is not what I am allowed to say.

What magnesium glycinate actually is

Magnesium is a reactive metal. It is never sold on its own, so every magnesium supplement is magnesium bound to a partner molecule, and that partner is the whole reason one product behaves differently from another.

In magnesium glycinate, the partner is glycine, the smallest of the amino acids. One magnesium ion sits between two glycine molecules, held by both the nitrogen and the oxygen on each one. That arrangement is called a chelate, from the Greek for claw, which is a fair description of what it looks like.

The chemistry is not in dispute. PubChem lists it as C4H8MgN2O4, molecular weight 172.42 g/mol, CAS number 14783-68-7, with the official synonyms magnesium diglycinate and bis(glycinato-N,O)magnesium. What magnesium does in the body

An adult body holds roughly 25 grams of magnesium. The US Office of Dietary Supplements fact sheet puts 50 to 60 per cent of that in bone, most of the rest in soft tissue, and less than 1 per cent in blood serum. That last figure is worth holding onto, because it is the reason a standard blood test is a poor guide to your magnesium status.

On what it does, the commonly quoted figure is 300 enzyme systems. A 2021 review in Nutrients went and counted what the enzyme databases actually list, and reported over 600 enzymes with magnesium as a cofactor, plus another 200 where it may act as an activator.

The single most important of those roles is unglamorous. ATP, the molecule your cells use to move energy around, is not biologically active on its own. It works as a magnesium complex. As that review puts it, magnesium acts as a cofactor in essentially all reactions involving the use and transfer of ATP. Magnesium is not an energy supplement in the way a coffee is, but energy metabolism does not run without it.

The glycine half of the molecule

Glycine is an amino acid your body makes for itself and also gets from food, particularly from collagen rich foods like slow cooked meat, skin and bone broth. A 2013 review in Amino Acids classifies it as conditionally essential, meaning the body can make it but not always in the quantities it needs.

In a magnesium glycinate capsule, though, glycine is not there as a second active ingredient. The amount is small, and it is there to carry the magnesium. That is a distinction worth being clear about, because a lot of marketing quietly credits magnesium glycinate with everything glycine has ever been studied for.

What the glycine does change is the route in, which is the next section.

How much of it you actually absorb

Magnesium gets across the gut wall two ways. Jahnen-Dechent and Ketteler, writing in Clinical Kidney Journal, describe the bulk of it moving passively between the cells, driven by concentration gradient and solvent drag, with a smaller active route through the TRPM6 and TRPM7 channels. The small active route is the one your body regulates, and it works hardest when your magnesium is low.

The practical consequence of that design is the single most useful thing to understand about magnesium dosing. The classic measurement comes from a 1991 study at the University of Texas Southwestern, published in the Journal of Clinical Investigation, which fed healthy volunteers five different magnesium loads and measured what got in.

As the dose went up, the proportion absorbed went down. Fractional absorption fell progressively from 65 per cent at the lowest intake to 11 per cent at the highest.

You still absorb more in absolute terms from a bigger dose. You just absorb far less of it, and the remainder stays in the gut, which is where the digestive complaints come from. Across the ordinary diet, the Office of Dietary Supplements puts typical absorption at 30 to 40 per cent.

The glycinate form has one piece of direct human evidence behind its reputation, and it is worth reading honestly. A double blind randomised crossover trial published in JPEN gave 12 patients who had undergone ileal resection a 100 mg dose of isotope labelled magnesium, as either the glycinate chelate or magnesium oxide. Across the group as a whole there was no difference, 23.5 per cent against 22.8 per cent. In the four patients whose absorption was most impaired, the chelate did considerably better, 23.5 per cent against 11.8 per cent, it appeared in the blood significantly sooner, and it was better tolerated by every patient. The authors concluded that part of the chelate is likely absorbed intact through a dipeptide transport pathway rather than as a free mineral.

How much of it is actually magnesium

This is where labels get slippery. Magnesium makes up 24.3 of the 172.4 units of molecular weight in anhydrous magnesium glycinate, which is 14 per cent. The other 86 per cent is glycine.

In practice the raw material used in manufacturing carries water as well, so the real figure on a finished product is usually closer to 11 or 12 per cent. The number to compare between products is the elemental one, never the compound weight. A tub shouting 500 mg on the front may be delivering 60 mg of magnesium. How people actually take it

Three patterns come up over and over in what customers tell us, and each one has at least a defensible reason behind it.

In the evening. Most magnesium glycinate products, ours included, direct you to take it at night. Being straight about the reasoning: that is convention and customer preference as much as anything. Magnesium is not a sedative, and the research on magnesium and sleep is modest rather than dramatic.

Every day, not now and then. Magnesium is a nutrient rather than a drug. There is no loading phase and nothing to feel in the first hour. What you are doing is topping up an intake, and intake is a daily number.

Split, if the dose is large. This one has the strongest rationale of the three. Given how sharply fractional absorption falls with dose size, two smaller doses will generally be better absorbed and better tolerated than one large one. If a product gives you 400 mg of elemental magnesium in a single hit, splitting it is sensible.

With or without food is genuinely unsettled. Food slows things down and some people find it easier on the stomach. If your product does not specify, either is reasonable. We go into the timing question properly in when to take magnesium glycinate.

Dose, and the ceiling worth knowing

The Australian and New Zealand Nutrient Reference Values set the recommended dietary intake at 310 mg a day for women aged 19 to 30 and 320 mg from 31, and 400 mg then 420 mg for men. That is total intake from all sources, and most of it is meant to come from food. Green leafy vegetables, legumes, nuts, seeds and wholegrains are the usual suspects.

The number people miss sits underneath: an upper level of 350 mg per day from supplements specifically. It does not apply to magnesium in food, because the kidneys manage dietary magnesium comfortably. Supplemental magnesium arrives all at once and behaves differently. A serve delivering 100 to 200 mg of elemental magnesium sits well inside that, and anything pushing 400 mg in one serve deserves a second look.

The bottom line

Magnesium glycinate is magnesium carried by two molecules of glycine. The chelate appears to be partly absorbed through an amino acid route rather than purely as a free mineral, which is the mechanism behind its reputation for being easier on the stomach, and the human evidence for that reputation is real but narrower than the marketing suggests.

The trade off is elemental content. You need more compound to deliver the same magnesium, so check that number on the label rather than the big one on the front.

Most products direct evening use and that is what the majority of people do, but there is no strong evidence that timing changes how much you absorb. Consistency matters more than the hour. Follow the directions on your product.

Can you take too much magnesium?

The Australian and New Zealand Nutrient Reference Values set an upper level of 350 mg a day from supplements specifically, separate from what you get in food. Going over it is the usual cause of digestive upset. Anyone with reduced kidney function should talk to their doctor before supplementing at all.

Does a blood test show if I am low in magnesium?

Less than 1 per cent of the body's magnesium is in blood serum, and the body works to keep that fraction stable, so a standard serum test is an insensitive measure of total body status. Interpreting one is a job for your doctor.
